Scientists can also see the terrible effects of Alzheimer's disease when they look at brain tissue under the microscope. Scientists are not absolutely sure what causes cell death and tissue loss in the Alzheimer's brain, but the plaques and tangles in the figures below are prime suspects.

The most damaging form of beta-amyloid may be groups of a few pieces rather than the plaques themselves. The small clumps may block cell-to-cell signaling at synapses. They may also activate immune system cells that trigger inflammation and devour disabled cells.

In mild to moderate stages, brain regions important in memory and thinking and planning develop more plaques and tangles than were present in early stages. As a result, individuals develop problems with memory or thinking serious enough to interfere with work or social life. They may also get confused and have trouble handling money, expressing themselves and organizing their thoughts. Many people with Alzheimer's are first diagnosed in these stages.

This question has been the focus of drug development for many years. BrightFocus has funded the work of scientists who have shown various methods of clearing away plaques. For example, Dr. Matthew Campbell (Trinity College) has experimented with temporarily disrupting the blood-brain barrier to clear out existing plaques. After many heartbreaking clinical trial failures, there is growing concern among the scientific and medical communities that clearing plaques may not be sufficient to treat the disease. This is a very active area of research.

The presence of plaques around a neuron causes them to die, possibly by triggering an immune response in the immediate area. Tangles form inside of neurons and interfere with the cellular machinery used to create and recycle proteins, which ultimately kills the cell.

Amyloid plaques appear early during Alzheimer's disease (AD), and their development is intimately linked to activated astrocytes and microglia. Astrocytes are capable of accumulating substantial amounts of neuron-derived, amyloid beta(1-42) (Abeta42)-positive material and other neuron-specific proteins as a consequence of their debris-clearing role in response to local neurodegeneration. Immunohistochemical analyses have suggested that astrocytes overburdened with these internalized materials can eventually undergo lysis, and radial dispersal of their cytoplasmic contents, including Abeta42, can lead to the deposition of a persistent residue in the form of small, GFAP-rich, astrocytic amyloid plaques, first appearing in the molecular layer of the cerebral cortex. Microglia, most of which appear to be derived from blood monocytes and recruited from local blood vessels, rapidly migrate into and congregate within neuritic and dense-core plaques, but not diffuse plaques. Instead of internalizing and removing Abeta from plaques, microglia appear to contribute to their morphological and chemical evolution by facilitating the conversion of existing soluble and oligomeric Abeta within plaques to the fibrillar form. Abeta fibrillogenesis may occur largely within tiny, tube-like invaginations in the surface plasma membrane of microglia. These results highlight the therapeutic potential of blocking the initial intracellular accumulation of Abeta42 in neurons and astrocytes and inhibiting microglia-mediated assembly of fibrillar Abeta, which is particularly resistant to degradation in Alzheimer brain.

Psoriasis is an autoimmune condition that causes inflammation in your skin. Symptoms of psoriasis include thick areas of discolored skin covered with scales. These thick, scaly areas are called plaques.

Psoriasis and eczema are two different skin conditions. Both conditions cause similar symptoms like discolored skin, a rash and itching. Psoriasis plaques cause areas of thick skin covered in scales. Eczema causes a rash of dry and bumpy skin. Eczema also typically causes more intense itching than psoriasis.

If you have psoriasis, your immune system is supposed to destroy foreign invaders, like bacteria, to keep you healthy and prevent you from getting sick. Instead, your immune system can mistake healthy cells for foreign invaders. As a result, your immune system creates inflammation or swelling, which you see on the surface of your skin as skin plaques.

It usually takes up to 30 days for new skin cells to grow and replace old skin cells. Your over-reactive immune system causes the timeline of new skin cell development to change to three to four days. The speed of new cells replacing old cells creates scales and frequent skin shedding on top of skin plaques.

Digging deeper, they traced the dense-core plaques using live imaging. Much to their surprise, the team discovered that after a microglial cell eats a diffuse plaque, it transfers the engulfed amyloid-beta to a highly acidic compartment and converts it into a highly compacted aggregate that is then transferred to a dense-core plaque. The researchers propose that this is a beneficial mechanism, organizing diffuse into dense-core plaque and clearing the intercellular environment of debris.

The Pioneer plaques are a pair of gold-anodized aluminum plaques that were placed on board the 1972 Pioneer 10 and 1973 Pioneer 11 spacecraft, featuring a pictorial message, in case either Pioneer 10 or 11 is intercepted by intelligent extraterrestrial life. The plaques show the nude figures of a human male and female along with several symbols that are designed to provide information about the origin of the spacecraft.[1]

The Pioneer 10 and 11 spacecraft were the first human-built objects to achieve escape velocity from the Solar System. The plaques were attached to the spacecraft's antenna support struts in a position that would shield them from erosion by interstellar dust.

Both Pioneers 10 and 11 have identical plaques; however, after launch, Pioneer 11 was redirected toward Saturn and from there it exited the Solar System. In this regard the Pioneer 11 plaque is somewhat inaccurate.The Saturn flyby of Pioneer 11 would also greatly influence its future direction and destination as compared to Pioneer 10, but this fact is not depicted in the plaques.

The research is focused on a protein called amyloid-beta. The protein normally carries out its functions in the brain in a form that is soluble, meaning dissolvable in water, but it sometimes hardens into clumps, known as amyloid plaques.

Previous research from the team found that regardless of the buildup of plaques in the brain, people with high levels of soluble amyloid-beta were cognitively normal, while those with low levels of the protein were more likely to have cognitive impairment.

The research found that with a baseline level of soluble amyloid-beta in the brain above 270 picograms per milliliter, people can remain cognitively normal regardless of the amount of amyloid plaques in their brains.

Espay said it will be important to ensure that the elevated levels of the protein introduced into the brain do not then turn into amyloid plaques, since the soluble version of the protein is needed for normal function to make an impact in the brain.
